1、大鼠局灶性腦缺血后下丘腦pSTAT3陽(yáng)性細(xì)胞的分布和局部微血管構(gòu)筑的關(guān)系1第四軍醫(yī)大學(xué)西京醫(yī)院麻醉科 2 第四軍醫(yī)大學(xué)基礎(chǔ)部神經(jīng)科學(xué)研究所雷翀1 成丹丹1 王百忍2 沈?qū)W峰2 熊利澤1摘要 目的 探討局灶性腦缺血后下丘腦內(nèi)磷酸化信號(hào)轉(zhuǎn)導(dǎo)與轉(zhuǎn)錄激活因子3（pSTAT3）免疫陽(yáng)性細(xì)胞的分布與局部微血管構(gòu)筑的關(guān)系。方法 雄性SD大鼠，隨機(jī)分成假手術(shù)組（n=8）和腦缺血組(n=8) 。腦缺血組采用線栓法致大腦中動(dòng)脈阻閉（MCAO，120min），假手術(shù)組未行MCAO。缺血再灌注24h后，每組各3只動(dòng)物用多聚甲醛灌流固定，用免疫組化法顯示下丘腦內(nèi)的pSTAT3陽(yáng)性細(xì)胞。每組各5只動(dòng)物用單寧酸-氯化鐵灌注

2、法媒染，以顯示腦內(nèi)各級(jí)血管。用免疫組化法在部分切片上顯示pSTAT3陽(yáng)性細(xì)胞，觀察陽(yáng)性細(xì)胞和血管分布的關(guān)系。結(jié)果 缺血再灌注24h后，缺血組動(dòng)物的神經(jīng)功能障礙評(píng)分顯著高于假手術(shù)組（P40.7)個(gè)。單寧酸-氯化鐵媒染顯示血管豐富的下丘腦區(qū)域，pSTAT3免疫反應(yīng)陽(yáng)性細(xì)胞數(shù)量也較多，且多位于血管附近。結(jié)論 腦局灶性缺血再灌注后，pSTAT3陽(yáng)性細(xì)胞的分布與局部腦血管的構(gòu)筑有一定的關(guān)系。關(guān)鍵詞 腦缺血；再灌注損傷；STAT3；血管構(gòu)筑Topographical relation between the distribution of phosphorylated STAT3-immunoreacti

5、domly divided into two groups (n=8 each), the sham-operated group and the focal cerebral ischemia-reperfusion group. The model of focal cerebral ischemia was established by intraluminal thread occlusion of the middle cerebral artery for 120 min. Twenty-four hours after reperfusion, animals were fixe

6、d by perfusion of 4% paraformaldehyde through the heart. The brains of three animals in each group were sectioned and processed for immunohistochemistry to examine the distribution of pSTAT3-like immunoreactive cells in the hypothalamus. Another 5 rats in each group were perfused by using tannic aci

7、d-ferric chloride method (TA-Fe) to stain the microvessel in the brain and immunocytochemistry technique was used as well to demonstrate the pSTAT3- immunoreactive cells in the slices. Results: At 24h after reperfusion, the neurologic deficit scores in ischemia group were significantly higher than t

8、hat in sham-operated group (P=0.000). In the former group, numerous pSTAT3-immunoreactive cells were seen in the hypothalamus ipsilateral to ischemia. The positive cells distributed mainly in tissue band beneath the pia mater, periventricular region, rostral portion of medial hypothalamic area (828.

9、872.7), paraventricular hypothalamic nucleus (1078.538.6) and supraoptic nucleus (361.840.7). The distribution pattern of pSTAT3-immunoreactive cells in the hypothalamus was coincidence with the density of microvessel in the region. More pSTAT3-immunoreactive cells were seen in the area where the microvessels were abundant. Furthermore, many positive cells located in close vicinity to the vessels. Conclusion: The distribution pattern of the pSTAT3-immunoreactive cells is related to the angioarchite
