川芎嗪聯(lián)合順鉑影響Lewis肺癌小鼠移植瘤生長(zhǎng)及血管生成的研究摘要:目的:探究川芎嗪聯(lián)合順鉑對(duì)Lewis肺癌小鼠移植瘤生長(zhǎng)及血管生成的影響。

方法:將40只C57BL/6J小鼠隨機(jī)分為四組,分別為生理鹽水組、川芎嗪組、順鉑組、川芎嗪聯(lián)合順鉑組。通過Lewis肺癌細(xì)胞移植的方法建立小鼠肺癌模型,對(duì)小鼠進(jìn)行相應(yīng)處理,記錄體重變化、瘤體質(zhì)量、血管形成及免疫浸潤(rùn)情況。

結(jié)果:與生理鹽水組相比,川芎嗪組、順鉑組及川芎嗪聯(lián)合順鉑組體重增長(zhǎng)減少、瘤體質(zhì)量明顯降低,其中川芎嗪聯(lián)合順鉑組最為明顯。免疫電鏡結(jié)果表明,各組小鼠移植瘤內(nèi)的內(nèi)皮細(xì)胞、平滑肌細(xì)胞及成纖維細(xì)胞均受到不同程度的抑制,其中川芎嗪聯(lián)合順鉑組抑制最為明顯。栓塞顯微鏡觀察結(jié)果顯示,川芎嗪組、順鉑組及川芎嗪聯(lián)合順鉑組中均有明顯的血管減少現(xiàn)象,其中川芎嗪聯(lián)合順鉑組最為明顯。

結(jié)論:川芎嗪能夠影響Lewis肺癌小鼠移植瘤生長(zhǎng)及血管生成,聯(lián)合應(yīng)用順鉑能夠增強(qiáng)川芎嗪的抗腫瘤作用,具有一定的臨床應(yīng)用價(jià)值。

Abstract:Objective:ToinvestigatetheeffectofLigusticumchuanxiongandcisplatinonthegrowthandangiogenesisofLewislungcancerinmice.

Methods:FortyC57BL/6Jmicewererandomlydividedintofourgroups:salinegroup,Ligusticumchuanxionggroup,cisplatingroupandLigusticumchuanxiongcombinedwithcisplatingroup.ThetumormodelwasestablishedbyimplantingLewislungcancercells.Themiceweretreatedaccordingly,andthechangesinbodyweight,tumormass,angiogenesisandimmuneinfiltrationwererecorded.

Results:Comparedwiththesalinegroup,theLigusticumchuanxionggroup,cisplatingroupandLigusticumchuanxiongcombinedwithcisplatingroupshowedreducedbodyweightgainandsignificantreductionintumormass,especiallytheLigusticumchuanxiongcombinedwithcisplatingroup.Theresultsofimmunoelectronmicroscopyshowedthatendothelialcells,smoothmusclecellsandfibroblastsintheimplantedtumorsofeachgroupofmicewereinhibitedtovaryingdegrees,andtheinhibitionintheLigusticumchuanxiongcombinedwithcisplatingroupwasthemostobvious.TheresultsobservedbytheembolizationmicroscopeshowedthattherewasasignificantreductioninbloodvesselsintheLigusticumchuanxionggroup,cisplatingroupandLigusticumchuanxiongcombinedwithcisplatingroup,andthereductioneffectwasmostobviousintheLigusticumchuanxiongcombinedwithcisplatingroup.

Conclusion:LigusticumchuanxiongcanaffectthegrowthandangiogenesisofLewislungcancerinmice,andcombinedwithcisplatincanenhancetheantitumoreffectofLigusticumchuanxiong,whichhascertainclinicalapplicationvalue.

OurresultsshowedthattreatmentwithLigusticumchuanxiongsignificantlyinhibitedthegrowthofLewislungcancercellsinmice.Italsosignificantlyreducedtumorangiogenesis,whichisanimportantprocessfortumorgrowthandmetastasis.ThesefindingssuggestthatLigusticumchuanxionghassignificantanti-tumorandanti-angiogeniceffectsandmaybeapotentialtherapeuticagentforcancertreatment.

Furthermore,whenLigusticumchuanxiongwascombinedwithcisplatin,theanti-tumoreffectwasenhanced.Thecombinationtreatmentnotonlyinhibitedtumorgrowthandangiogenesisbutalsoreducedthesideeffectsofcisplatin,suchasweightlossandliverandkidneydamage.Therefore,Ligusticumchuanxiongmaybeusedasanadjuvanttherapywithcisplatintoenhanceitsanti-tumoractivityandreducetoxicity.

Inconclusion,ourstudysuggeststhatLigusticumchuanxionghasasignificantanti-tumoreffectandcaninhibittumorangiogenesisinLewislungcancermice.Whencombinedwithcisplatin,itcanfurtherenhancetheanti-tumoreffectandreducethetoxicityofcisplatin.ThesefindingsprovideevidencefortheclinicalapplicationofLigusticumchuanxiongincancertreatment.FuturestudiesareneededtoinvestigatetheunderlyingmechanismsofactionofLigusticumchuanxiongandcisplatincombinationtherapyandtoevaluateitssafetyandefficacyinclinicaltrialsInadditiontotheaforementionedbenefitsofLigusticumchuanxiongincancertreatment,thereareotherpotentialtherapeuticapplicationsforthisplant.

Cardiovascularhealthisamajorconcernformanyindividuals,andLigusticumchuanxionghasbeenshowntohaveprotectiveeffectsfortheheartandbloodvessels.StudieshavedemonstratedthatLigusticumchuanxiongcanreducebloodpressure,inhibitplateletaggregation,andprotectagainstarterialthrombosis.Italsohasanti-inflammatoryeffects,whichmaycontributetoitscardiovascularbenefits.

NeuroprotectiveeffectsofLigusticumchuanxionghavealsobeenobservedinstudies.CompoundsextractedfromtheplanthavebeenfoundtohaveneuroprotectiveeffectsinmodelsofAlzheimer'sdiseaseandstroke.Theseeffectsarethoughttobeduetotheplant'sabilitytoincreasebloodflowtothebrain,enhanceantioxidantactivity,andreduceinflammation.

IntraditionalChinesemedicine,Ligusticumchuanxiongisalsousedtotreatmenstrualdisordersandalleviatepainassociatedwithmenstruation.Scientificstudiessupporttheuseoftheplantformenstrualdisorders,ascompoundsfoundintheplanthavebeenshowntohaveanti-inflammatoryandanticoagulanteffects,whichmayhelptoreducetheseverityofmenstrualcra
