Abstract:

Thispaperisabouttheautomaticnucleicacidextraction/amplificationmethodandinstrumentbasedonthewater-oilsystemandMagPack.Theaimofthisstudyistoenhancetheautomationofnucleicacidextractionandthesensitivityandaccuracyofnucleicaciddetection.Firstly,westudiedthewater-oilsystemandMagPack,anddeterminedtheoptimalextractionconditionsandamplificationscheme.Secondly,wedesignedanautomaticnucleicacidextraction/amplificationinstrumentandconductedexperimentstoverifyitsperformance.Theresultsshowedthattheinstrumentcouldextractandamplifynucleicacidquicklyandaccurately,withgoodreproducibilityandrepeatability.Thisstudyprovidesanewautomatedmethodandinstrumentfornucleicaciddetection,whichhasgreatpotentialforapplication.

Inthisstudy,theresearchersdevelopedanewautomationinstrumentfornucleicacidextractionandamplificationbasedonthewater-oilsystemandMagPacktechnology.TheMagPacktechnologyusesmagneticparticlesascarrierstoextractorpurifynucleicacidmoleculesfrombiologicalsamples.Thewater-oilsystemisauniquetechnologythatseparatesthenucleicacidsfromthesamplebyimmersingitinanaqueousphaseandanon-miscibleorganicphase.
