轉(zhuǎn)錄因子SNAIL在小鼠植入前胚中的表達(dá)及作用摘要:轉(zhuǎn)錄因子SNL是一種能夠促進(jìn)細(xì)胞上皮間質(zhì)轉(zhuǎn)化(EMT)過(guò)程的關(guān)鍵蛋白質(zhì),其在胚胎發(fā)育和腫瘤轉(zhuǎn)移中起著重要作用。本研究通過(guò)實(shí)時(shí)熒光定量PCR、免疫熒光染色和Westernblot等方法對(duì)小鼠植入前胚中SNL轉(zhuǎn)錄因子的表達(dá)和作用進(jìn)行了探究。結(jié)果表明,在小鼠植入前胚中SNL表達(dá)水平較低,但仍然存在一定的表達(dá)及調(diào)控作用。通過(guò)通過(guò)SiRNA技術(shù)或過(guò)表達(dá)實(shí)驗(yàn),發(fā)現(xiàn)SNL在控制小鼠植入前胚細(xì)胞上皮細(xì)胞的數(shù)量和形態(tài)方面扮演著重要角色,同時(shí)還可能參與小鼠胚胎早期器官形成。該研究對(duì)于深入了解轉(zhuǎn)錄因子SNL在小鼠植入前胚中的作用機(jī)制,以及研究動(dòng)物胚胎發(fā)育和疾病的發(fā)生機(jī)制具有一定的價(jià)值。

Abstract:TranscriptionfactorSNLisakeyproteinthatpromotestheprocessofepithelial-mesenchymaltransition(EMT).Itplaysanimportantroleinembryonicdevelopmentandtumormetastasis.Inthisstudy,theexpressionandfunctionofSNLtranscriptionfactorsinmouseblastocystswereinvestigatedusingreal-timequantitativePCR,immunofluorescencestaining,andWesternblotting.TheresultsshowedthatSNLexpressioninmouseblastocystswaslowbutstillhadacertainlevelofexpressionandregulation.ThroughSiRNAtechnologyoroverexpressionexperiments,itwasfoundthatSNLplayedanimportantroleincontrollingthenumberandmorphologyofepithelialcellsinmouseblastocystsandmayalsoparticipateinearlyorganformationinmouseembryos.ThisstudyisofgreatvalueforadeeperunderstandingoftheroleofSNLtranscriptionfactorsinmouseblastocystsandforstudyingthemechanismsofanimalembryonicdevelopmentanddisease.

Finally,thestudyofSNLhighlightstheimportanceoftranscriptionfactorsinregulatinggeneexpressionandcellulardifferentiation.Inrecentyears,therehasbeengrowinginterestinusingtranscriptionfactorstomanipulatecellfateinvariousapplications,suchasregenerativemedicineandtissueengineering.ByunderstandinghowtranscriptionfactorslikeSNLinteractwiththegenomeandothercellularcomponents,researcherscoulddesignmoreefficientandprecisemethodsfordirectingcelldifferentiationandcreatingfunctionaltissues.
